What would cause 'lock tables' to stop working?

I upgraded my site to the latest version earlier tonight, and now my comments are generating the 'lock tables' SQL error (yes, I've read that page).
Everything was working fine until I upgraded, and now comments are broken. Any ideas why comments have gone south? Any help would be appreciated.
