The long answer is yes, a plugin may affect page creation times since some plugin code will be run on every page load and depending on how the plugin is implemented it may affect page load times.

Depends on what the plugin does.
Every plugin will cause a (very) slight slowdown, as it means more files will have to be loaded for every page (the functions.inc, config.php, possibly language files). That shouldn't really be measurable, though, normally.
Try disabling the plugin and see if it makes a difference.
